Confocal microscopy offers several advantages over conventional optical microscopy, including controllable depth of field, elimination of image degrading out-of-focus information, collection of serial optical sections and reconstruction of cells in tridimensional view.
The presentation will focus on applications in confocal microscopy available on the CRP-Santé platform. After a short introduction to confocal microscopy I will present different examples of experiments to illustrate the possibilities of the system. The first part will focus on one of the major application; imaging cells in 3D. We will see the different solutions in terms of fluorescence labelling (immunolabelling, fluorescent probes and fluorescent proteins), and how to acquire and visualize your datasets. Different software solutions are available to reconstruct, animate and present your results. The second part will concern live cell imaging and dynamic information that can be obtained from timelapse experiments. Then the third part will be dedicated to image analysis, we will see what kind of data can be extracted from the pictures and how to get qualitative and quantitative measurements.
